Digestion :
- 8C/14K, 18A/24A (2 clones :one ligation of the 19/08, one of the 07/09) : with E+P
- 10L, 24A, I0500, 4O : with E
Verification of the digestion with an Agarose Gel electrophoris :
- 10L, 4O : they present the same bands on the wrong weight, maybe contamination
- I0500 : no DNA
- 24A : ok
Culture on plate of 4O/10L (ligation of the 13/09), 14K, I0500 and 5 new clones of phaA/phaB (ligation of 07/09)
Ligation of 1F/24A
First purification of phasin-intein
Measure of fluorescence and DO on the PHL1273 cultures
