Today the work wa focused on the digestion reaction of three parts, RBs, Promotor and the plasmid.
1) Promotor
Buffer 2 ------------> 5 ul
BSA -----------------> 0,5 ul
EcoR1----------------> 1 ul
Spel ----------------> 1 ul
DNA(BBa_1719005)-----> 20 ul
H2O -----------------> 22,5 ul
Final volume= 50 ul.
2) RBS
Buffer 2 ------------> 0,5 ul
BSA -----------------> 5 ul
Xba -----------------> 1 ul
Pst1 ----------------> 1 ul
DNA(BBa_B0034)-------> 20 ul
H2O -----------------> 22,5 ul
Final volume= 50 ul.
3) Plasmid
Buffer 2 ------------> 0,5 ul
BSA -----------------> 5 ul
EcoR1----------------> 1 ul
Spel ----------------> 1 ul
DNA(BBa_psB1A2)------> 20 ul
H2O -----------------> 22,5 ul
Final volume= 50 ul.
At the end we do the electroforesis in gel and all the parts were digested well. =)
