11 clones appear for the pha A/pha B ligation of the 19/09. Start of a culture on plate for each
Agarose gel electrophoresis for verification :
- 8C is OK
- 8C/14K and 4O/10L : not OK
Liquid cultures of 8C/14K (2 other clones), Curli/22B and 1F/24A
Measure of fluorescence and DO of the liquid culture of PHL1273 of the 20/09. Start of 6 new cultures :
- 3 in a water bath at 33°C, shaking speed 100 rpm
- 3 in a water bath at 34°C, shaking speed 100 rpm
Agarose gel for ompR234 PCR verification (0,7%): OK, one major strip for both dilutions between 1000bp and 750bp (expected size of ompR234: 790bp). Negative control is OK.
