Measure of the fluorescence and DO of the cultures of PHL1273
Start of 6 cultures of PHL1273 and 2 of PHL818 (the same as PHL1273 without the plasmid containing Curli promoter and unstable GFP).Two conditions of culture (with 3 tubes of PHL1273 and one of PHL818 for each) :
- waterbath at 29°C, shaking speed 100 rpm
- waterbath at 30°C, shaking speed 100 rpm
Some clones appear for the ligation Curli/22B (14/09) and 1F/24A (15/09)
DNA extraction of 4O, I0500, 14K, phaA/phaB (clones 11 to 15)
Liquid cultures of : 18A, L2/1N, 18A/24A, Phasin-intein, phasin, Curli/22B
