Results of the Transformation of 1F/24A: about a hundred of clones.
We select 10 clones and we make a liquid pre culture with Amp.
New digestion of 1F by S+P and 24A by X+P (extracted on 07/27)
Agarose gel electrophoresis: ok
Extraction and purification of the band 24A with a kit (concentrated in 20µL)
Ligation of 1F/24A: ratio--> 5/10µL
Transformation of the ligation
Transformation of L3 on tetracycline plate
