Monday 8/23

• Ligate L90, L91, and L92 (overnight ligation)

Tuesday 8/24

• Transform the ligation product from yesterday

Wednesday 8/25

• Pick colonies from the transformed cells. Perform colony PCR and run gel on the PCR product.
  • Gel shows negative result Failed
• Set up HSP-LacI-GFP experiment with glucose concentration (.1mM, .5mM, 1mM, 5mM)

Thursday 8/26

• Glucose appears to not effect the GFP expression in this experiment which contradicts the result from previous experiment.

Friday 8/27

Grow up cell cultures, redesign Glucose-GFP expression experiment, and prepare to run the new experiment for monday.
