Parts Assembly

Colony PCR of the overnight grown colonies from 15.10.2010 was performed. twenty four colonies for each part were picked. The PCR samples were run on an agarose gel and positive clones were identified and kept for overnight incubation.

Fusion Protein

Colony PCR was done to the transformed ligations and many colonies were chosen for the overnight culture. Also colony PCR was done to the ligated fusion protein into the pETMM43 Gradient PCR for the MBP from pETMM43 and choosing the ideal temperature for its amplification
