The overnight ligation samples did not have any colonies grown on them. The 20f and 21f of the first ligation showed some colonies and hence were used for colony PCR. Hence the following parts were subjected to colony PCR:

4a, 9b, 13e, 15e, 14f, 20f, 21f, 10, 11, 42, 43
