Gel of pGEM+WillRS digest

Lanes 1-4 are pGEM+WillRS lig. digest with BamHI+NcoI, buffer3

Gel isolated with qiagen kit, eluted in 45ul buffer EB

M9 minimal media

M9 salts 5x

  • In 1ul dH2O
    • 30g Na2HPO4
    • 15gKH2PO4
    • 5g NH2Cl
    • 2.5g NaCl
    • 15mg CaCl2 (optional, added this time)
  1. Dilute to 1x: take 200mL M9 salts and add dH2O to final vol. of 1L
  2. Autoclave
  3. Add the following after autoclaving (per L)
    • 1mL 1M MgSO4*7H2O
    • 20mL 20% carbon source (dextrose)
    • 5mL 20% casamino acids
    • 0.1mL 0.5% thiamine

Notes - will need to filter sterilize. Add antibiotics as needed before use
