Transformation Results
- RFP control: 100+ colonies
- Ligation control: 200 colonies, 14 were blue
- Mnt-pGEM: 1 colony out of 30 was white. Placed into LB/Amp 4mL liquid culture
- CI-pGEM: 8 colonies, all blue. Placed back in incubator
- AraC-pGEM: 5 colonies, all blue. Placed back in incubator
Retransformed CI and AraC + pGEM into XL1Blues on Amp+ LB plates w/ xGAL and IPTG for blue white selection
Re-ligation of AraC and CI into pGEM
This time, we are increasing the amount of insert DNA
50ul total volume
- 15ul ligase buffer 2x
- 2ul T4 ligase
- 1ul pGEM
- 12ul insert (CI or AraC)
