Culture

Materials

You will need:

  • Luria broth
  • Pasteur pipette
  • Pipetting Aid
  • Sterilized polypropylene tubes
  • Microtiter pipette & small sterilized pipette tips
  • Antibiotic resistance stock
  • Plates with the desired microbe to be cultured

Procedure

  • Light bunsen burner.
  • Pour 4 mL of luria broth into each polypropylene tube.
  • Pipette the required amount of antibiotic resistance into each tube. Recommended amounts can be looked up on openwetware.org.
  • Use a small pipette tip to lightly touch one isolated colony, then place into the respected tube.
  • Close tubes and then incubate at 37°C for approximately 12 to 16 hours.

Purpose

To grow the desired microbe in slightly larger quantities.
