Materials
You will need:
- Luria broth
- Pasteur pipette
- Pipetting Aid
- Sterilized polypropylene tubes
- Microtiter pipette & small sterilized pipette tips
- Antibiotic resistance stock
- Plates with the desired microbe to be cultured
Procedure
- Light bunsen burner.
- Pour 4 mL of luria broth into each polypropylene tube.
- Pipette the required amount of antibiotic resistance into each tube. Recommended amounts can be looked up on openwetware.org.
- Use a small pipette tip to lightly touch one isolated colony, then place into the respected tube.
- Close tubes and then incubate at 37°C for approximately 12 to 16 hours.
Purpose
To grow the desired microbe in slightly larger quantities.
