Retrieval of yesterday's transformation

Retrieved transforms 20h after incubation

  • Colonies grew to high density, some secondary colony growth?
  • No blue/white selection used to screen for insertion of Tat-L into pBS

Will redo transformation with 2ul of ligation product from 9/16

  • Add to2ul of ligation product
    • 50ul 20mM xGal
    • 2ul 1M IPTG
    • 100ul LB

Incubated at 3:30PM, plated all 300ul of mixture

To be out at 12pm, 9/19
