Results of blue-white selection for WillRS-pGEM ligation

We appear to have a good number of colonies (about 100-300), some white, some blue. No contamination.

  • Put in the fridge (4C) for 2 hours and then pick a colony

Plan: put into 3ul amp into 3mL cultures and grow out; miniprep all; digest and run on gel to verify which ones worked.

Made overnight culture of Lovtap r*2

MRS Broth-agar plates for miraculin expt.

  • 27.5g of mrs broth and 5g of bacto agar in 500ml water
  • Tet - 5ug/ml
    • 250ul of 10mg/ml stock in 500ml liquid
