Luria Broth

Materials

You will need:

  • Deionized water
  • Tryptone
  • NaCl
  • Yeast Extract

Procedure

  • Combine 10g tryptone, 5g yeast extract, 10g NaCl, and 950 mL of deionized water into a large beaker/Erlenmeyer flask.
  • Mix until dissolved.
  • Check to see if the pH is 7.0 using a pH meter. If needed, add 5N NaOH to adjust the pH to 7.0.
  • Fill solution to 1L with deionized water.
  • Aliquot into smaller glass bottles. (Optional)
  • Autoclave.

Purpose

For culturing desired microbes.
