Aim

To check if the digestion from yesterday worked.

Results

The result from gel electrophoresis:

Figure 1 shows the double digest of 12 tubes of pGFPrrnB and yneA.

• Lane 1: pGFPrrnB + yneA 1
• Lane 2: pGFPrrnB + yneA 2
• Lane 3: pGFPrrnB + yneA 3
• Lane 4: pGFPrrnB + yneA 4
• Lane 5: pGFPrrnB + yneA 5
• Lane 6: pGFPrrnB + yneA 6
• Lane 7: pGFPrrnB + yneA 7
• Lane 8: pGFPrrnB + yneA 8
• Lane 9: pGFPrrnB + yneA 9
• Lane 10: pGFPrrnB + yneA 10
• Lane 11: pGFPrrnB + yneA 11
• Lane 12: pGFPrrnB + yneA 12

Discussion

The bands we got from the gel shows that digestion in tubes 1, 2, 4, 10, 11 and 12 worked.

Conclusion

We use the digestion from the six tubes that worked for ligation.
