Wednesday Morning

Aims

The aim of today's Lab practice was to transform competent cells with GFP and RFP inserts in an RFP backbone.

Materials

• E.coli broth culture
• Pipettes
• Plasmid ligation products from yesterday
• Conical flasks

Protocol

Cells are made competent ready for transformation.

• Positive control(plasmid uncut)- to make sure cells are transfroming properly
• Negative control (just cells)- nothing but competent cells

5 microfuge tubes:

• 1:5
• 1:3
• Vector
• Negative add nothing
• Positive add 1 μl uncut

Wednesday afternoon - taking a break at the Hoppings!!!

Aims

The aim of this Lab practice was to have lots of fun!

Materials

• Candy floss
• Suntan lotion
• Teddy bears
• Burgers and bacon
• Chips and gravy
• Icecream
• Meerkat toys
• Madmouse ride!
• Beer, wine, lemonade
• Football
