プロジェクト

Dr. E. coli : 世界最小の注射器

   私たちのマインテーマは三型分泌装置という菌が持つ器官の中でも驚くべきものだと思います。この注射器に似た装置はタンパク質を菌の中から標的の真核細胞に打ち込むものです。しかしこの装置は病原性グラム陰性菌であるヤルシニアやサルモネラの器官です。私たちが非病原性の大腸菌を用いてそれを安全使えることを目指しました。

PCR用のアセンブリープロトコール

   時々、スタンダードプロトコールではプラスミドが増えなかったり、BioBrickがホストセルに有害で増幅されないことがある。こんな時にPCRはすごく役立つ。私たちはPCRを使った場合、制限酵素処理が合否がすぐ分かるように改善を加えました。

チーム

   北海道大学のigemのチームは日本で8番目につかれたチームです。一人のインストラクターと8人の学部生から構成されています。学部生の所属は理学部、医学部、農学部と様々です。今年、北大はチームはiGEM発出場である。

実験ノート

   成功と失敗の記録。失敗がたくさんあった道のりでしたがやってのけました。（実験ノートは英語でかかれています）

    私たちが使ったプロトコール集である。（現在英語、翻訳不定）

   私は本テーマに関連した2つのパーツを登録しました。一つ目はタンパク質につけ分泌シグナルで三型分泌装置に分泌させるためのもの。もう一方は三型分泌装置が正しく機能するかどうかを検証するためのレポーターである。

   またPCR用のアセンブリープロトコールに役立つプライマーセットも登録した。
