Retrieval of yesterday's transformation
Retrieved transforms 20h after incubation
- Colonies grew to high density, some secondary colony growth?
- No blue/white selection used to screen for insertion of Tat-L into pBS
Will redo transformation with 2ul of ligation product from 9/16
- Add the following to 2ul of ligation product:
- 50ul 20mM xGal
- 2ul 1M IPTG
- 100ul LB
Incubate at 3:30PM, plated all 300ul of mixture
To be out at 12pm, 9/19
