Quorum addiction module
- Inventory of all the parts already ready to use.
- Midiprep on the RBS strenght 1.
- Miniprep on some of the parts needed (AHL degrading enzyme, LuxR, Both promoters, LuxI and RBS strenght 0.3).
Module hydrogen
- After the PCR of verification, we found out that we obtained the ldhA deleted mutant. The other deletion candidate were all negative.
- We continue the purification process for the deletion candidate
- We continue to try to obtain other deletion candidates
- We tested the insertion of tdcE with the RBS.
Module homologous recombination
- We finally obtained the plasmid TOPO with the cm and kan resistance cassettes !
- We can now digest the two resistance cassette in order to insert them in the pSB1C3
Module heavy metals detection
- We inserted the gene of the carotene behind a constitutive promoter
- We inserted the copper sensitive promoter in front of a RFP
