Transformed trans1-T1 competent cells with the ligation products (24hours).

Spread the bacteria on plates with kanamycin and chloramphenicol. Also spread double resistance bacteria (Bacteria with T vectors harboring Chloramphenicol resistance gene) and bacteria harboring T vectors with scrambled sequence insertion.
