Emulsifier test

Materials:

- alkane

- 25 mM Tris buffer, pH 8

- emulsifier sample

- 0.1% Sudan II in 50% EtOH

- shaking incubator at 30 °C

- spectrophotometer


Protocol:

1. Determine protein concentration by Bradford assay

2. Mix, 0.02 mL of alkane (alkane = substrate), 0.1-0.5 mL of protein sample and Tris buffer to a total of ? mL in a cuvet

3. Add Tris buffer up to 1 mL

4. Add 20 μL 0.1% Sudan II

5. Vortex 15 seconds at max speed

6. Let sample rest for 1 minute

7. Dilute to get in the right absorbance range

8. Measure absorbance at OD493
