September 18th
DNA extraction :
- I0500 : 2 clones (plates from the 17/08 and 13/08)
- 1F/24A : 3 clones
- curli/22B :2 clones
- 4O/10L : 1 clone
PHL818 liquid culture in LB-Tet (GC) 8:45 AM - 11:15 AM: reaching a DO600nm = 0,8.
PHL818 culture ½ dilution (750µl culture + 750µl LB) and genomic DNA extraction on these 1,5ml with DNeasy Blood and Tissue Kit (QIAGEN). Note that 10µg of RNase A (Fermentas) had added and elution had been done with 200µl elution buffer, sample is finally stored at -20°C.
