Terminator leak switch assay

To realize 4C3 leak switch, we choose proper terminator which terminates transcription when connected two or more but leak when single.

We made assay (see Fig) to select proper terminator.

We modified this flpe by PCR:

1st -> cloning

2nd -> modified SpeI site

3rd -> modified EcoRI site

Anti-RNA repression assay

To get number information from same row, column and block, each E.coli have to repress transcription of unwanted virus RNA by antisense, we call "location sequence." (see Fig)

First, we order four location sequence and connect to rbs site. Then check the ability to stop transcription.
