Preparation

• ・Bacto trypton
• ・NaCl
• ・Bacto　Yeast
• ・MilliQ
• ・Agarose (for plate）

Procedure

100ml for liquid

• 1. Put substances below to the bottle.
  • 1g　Bacto trypton
  • 1g　NaCl
  • 0.5g　Bacto　Yeast
  • 100ml　MilliQ
• 2. Rap the bottle by aluminum foil, autoclave for 20 minutes at 120 degrees C.

100ml for plate（5 plates/100ml）

• 1. Put substances below to the bottle.
  • 1g　Bacto trypton
  • 1g　NaCl
  • 0.5g　Bacto　Yeast
  • 100ml　MilliQ
  • 1.5g　Agarose （for plate）
  • Antibiotics
• 2. Put in stirring bar, rap the bottle by aluminum foil, and autoclave for 20 minutes at 120 degrees C.
• 3. Add antibiotics after the bottle is cool enough to touch by hand.
• 4. Pour to the plate.
