Colony Pick Up

Preparation

  • test tube
  • LBbroth 4ml/tube
  • antibiotics

(take out from fridge and use after thawing out / needless to use after colony PCR)

Procedure

  • 1. add 2 μm of LB broth to the tube
  • 2. add the antibiotic to the tube(1μm for 1ml)
  • 3. poke the colony on the plate by toothpick(or tip of pipette)and put the toothpick to the tube
  • 4. label the cap of the tube and fix the cap on the tube
  • 5. incubate it at 37 degrees (shake culture)

(15~18 hours is best / 8 hours is the fastest)
