Proportional Calculation for repeating the cloning with pGEM-T Easy Vector System:

Insert:Vector molar ratios Recommended ratios: 3:1, 1:3

Vector (3kb) = 50 ng/ul

ng insert= (ng vector x kb insert/kb vector) x insert:vector molar ratio

50 ng vector x 2.2 kb insert/3 kb vector x 3:1 = 110 ng insert

110 ng = 1.03 ul of insert

Today we repeated the cloning and the ligation
