Making glycerol stocks

Materials

  • 1.5ml microcentrifuge tubes
  • Glycerol

Protocol

  1. Inoculate 5ml THY Broth with a single colony and incubate, stationary at 37°C for 8 hours.
  2. 0.5ml culture mixed with 0.5ml of sterile 50% glycerol (filter sterlise).
  3. Store at -80°C in microcentrifuge tubes.
