Transformation to E. coli

• Add 10 µL of ligate to 100 µL competent cells
• Put cells for 30 minutes on ice
• Heat shock the cells for 45 seconds at 42 °C
• Put cells on ice for 2 minutes
• Add 400 µL TY and grow for 1 hour at 37 °C
• Plate 100 µL of cells on LB agar (2%) plates
