Today we will be pouring new plates for streaking new transformed cells.

1) I located some 2% Ara/agur made by Greg M. on 4/10/10.

2) Place the 250 ml Elemyier flask in microwave to melt set on beverage 4oz setting and allow to run.

3) Locate protocol for adding amp. To the agur. from openwetware.org/wiki/ampicillin.

4) Hit run on microwave 4oz. bev. Setting for a sec. time

5) After melting agur we will add 1.16 mg/mL Ampicillin.

6) We added .6088 g Amp. powder to 5 mL of DI water then micropitetted 250mL into 20 centrifuge tubes freezing 19 of them in the -80 C freezer.

7) We added 250 mL to 250 mL LB agur when cooled then poured 16 plates labeling them LB/amp 8/24/10 J.H.

8) Let cool then place into bag and into fridge. --Rchamberlin 18:27, 30 September 2010 (UTC)
