THT staining


Thioflavin T is a stain that specifically binds to amyloid fibres. Since chaplins assemble into amyloids, this is a detection method worth trying. From literature we know that chaplins give emission between 460-600 nm with a peak at 482 nm. Samples are stained just before measurement in a spectrophotometer cuvette.


After TFA treatment samples are rediluted in demi water or tris buffer.


- Then vortex for two minutes to reassemble the chaplins into amyloids


Just before measurement with the spectrophotometer add 2 ul THT solution per ml sample and resuspend with the pipet.
