  • Transformed the ligated PyeaR parts
  • Miniprepped, digested, ran a gel electrophoresis (took only the bottom band (~3000bp), gel purified, dephosphorylated, and then PCR purified GFP
  • Ligated GFP to PalkB (the alkane promoter)
  • Incubated PyeaR+RFP overnight
