9/30/10

Today we have been looking at the IGEM registry to find a LacZ without GFP also with RBS and terminator and with the same assembly standard. I found a part ; BBa_I732017 we have it in our Distribution; Plate 2 well 3K So today I electorporated part I732017 into E.Coli bacteria . Electrocomp cells using protocol from page 15 I pulsed (2) twice 1st-1.8 kv at 5.5 ms 2nd=1.8 kv at 5.5 ms Place at 37 C for 1 hour then plate on agar/LB=lennox with amp.

NOTE: The experiment from Friday we are showing ver bright flourescen on part/trial F. We will perform the experiments again tomorrow

After waiding 45 minutes I removed from the incubator @ 37 C, the 1.5ml centrifuge tube containing our new sample of elctroporated E.Coli with part I732017 for the iegme Reg and too and cleaned the counter of workstation 4 and setup a spin streak setup shooting 100 ul of sample onto agar/amp/ara(only plate available), spun. I then placed it back into the incubator
