Monday 16th August - Standard

For most of the day Steve, Caz and Matt were autoclaving, pouring agar plates and preparing LB. Ampicillin and kanamycin plates were prepared separately and together. The restriction digest performed on the plasmid didn't work unfortunately, as the DNA ladder was too high in concentration compared to the palsmids.

Meanwhile Tom began profiling the team on the wiki and made 2 interviews, one with Groningen and the other with Harvard for human practices. Nii and Narmada tried to work out how a histidine kinase could be modelled by researching literature.


Tuesday 17th August - Re-restriction.

Steve, Caz and Matt attempted the restriction digest again with a lower concentration of ladder DNA compared to the plasmids. Tom returned home in the morning for a bit to carry on work on the wiki before returning at 2pm to interview DTU-Denmark. Nii and Narmada had a meeeting with Qaiser to try clarify some technical aspects of the modelling. Tom, Matt, Caz and Steve later plated out some cells to see if the transformations were successful on the Kanamycin and Ampicillin plates. At 5pm Tom interviewed Virginia.

Evening plans are at Matt's for homemade curry and Tekken!


Wednesday 18th August - A day for modelling.

Steve Wilkinson came to help Nii and Narmada by giving advice about how to carry out a function on matlab and to look at how they had modelled the system. The task for team lab was making loads of plates. Not much interesting to report frrom that aspect unfortunately, we're still waiting very patiently for our genes to arrive.


Thursday 19th August – Transformation.

The task today in labs was preparing transformations. Steve, Matt and Caz transformaed about 10 different biobricks in the competent cells which took up a lot of the day.

Team modelling tried to find out what was wrong with their code, as they didn't manage to get the correct sigmoidal curve. During the afternoon we all went down to Catherine's (MASSIVE) office to make some more progress on our collage. Only a little more to do after this and PLENTY of clearing up to do.


Friday 20th Sheffield - An achre of office to clean.

For most of the day we were in Catherine's office finishing off our identity collage. Lab work was also interspaced during the day to make another load of transformations, as all of yesterday's work was unsuccessful. The most important transformation of the day was with the pgA promoter.

Last thing left for the team to do was to clear up the whole load of dirty mugs, paper and crumbs we left all over Catherine's office. Let's hope she doesn't read this eh. Time for the weekend!
