Transformation to E. coli
- Add 10 µL of ligate to 100 µL competent cells
- Put cells for 30 minutes on ice
- Heat shock the cells for 45 seconds at 42 °C
- Put cells on ice for 2 minutes
- Add 400 µL LB and grow for 1 hour at 37 °C
- Plate 100 µL of cells on LB agar (2%) plates
