Today we redid the PCR for Pu since it did not work on Tuesday. One of the sets of primers worked. I am also attaching all of the gel pictures that we have so far. They are not great quality, so if you can't see something that should be there just email me back.
The pictures are named based on the date the lab was. Under this I am putting additional information about what each picture is showing (from the left side of the photo to the right side). They are all from gel electrophoresis.
8-17 There is a 2-log DNA ladder then a PyeaR band, it is correct
8-20 This is again a 2-log DNA ladder and then PyeaR, correct
8-20 (2) This is the double terminator and then the 1 kb DNA ladder (The gel messed up which is why the ladder is hard to read, but the terminator is probably correct)
8-23 This is the gel ran after the first Pu PCR attempt. There is a 1 kb DNA ladder (should have used the 2-log DNA ladder), then the Pu band, which looks correct
8-24 There is a band for Pu that is incorrect and then a 2-log DNA ladder
8-26 We tried four different combinations of primers for the Pu PCR and this is the gel with the results. First is the 2-log DNA ladder, then primers 1&2 incorrect, primers 1&4 maybe, primers 3&2 incorrect, and primers 3&4 correct.
