  • We digested PyeaR with the new SpeI and PstI-HF that we received in the mail.
  • We then ran a gel (see picture) which gave the correct band, we only took the bottom portion for gel purification since that is the part that is correct.
  • We then ligated PyeaR to GFP, RFP, and orange overnight at 14C.
