• Prepared glycerol stock of the transformed ligated parts, in case they are correct.
• Miniprepped the transformed ligated parts, and then digested it.
• We also digested another part using all the enzymes to test the enzymes, because we have gotten poor digestions in the past.
• We ran all the digestions on two gels. (pictures are attached)
• The first gel is the digested parts. They are not correct, therefore are ligation did not work. We will try again tomorrow.
• The second gel is the enzymes. The resulting gel was strange, especially the two middle results. This did not tell us much.
