  • Made chloramphenicol plates
  • Transformed the ligations from yesterday.
  • I also realized that the ligations will give us the wrong product. We took a random part (BBa_J04450) with the pSB1C3 backbone to digest to get just the backbone, but I did not digest it properly, so the ligations will give us our parts (PyeaR+GFP and PyeaR+RFP) behind LacI+RFP. Therefore I will be digesting and ligating again during the week.
