Today camera crew #2 came by from Nanopodium, a platform for exchanging thoughts, ideas, opinions and best practices on nanotechnology. They are making a movie about our project for the Nano Video Hunt. This is a campaign started with Hyves were young people are challenged to post videos about their views on nanotechnology.

I made some time for doing some transformations from the Biobrick plates, I will give you the list of the biobricks that I tried to transform.

# Code Resistance marker Function
1 J61100 Amp RBS Anderson family
2 J61101 Amp RBS Anderson family
3 J61107 Amp RBS Anderson family
4 J61117 Amp RBS Anderson family
5 J61127 Amp RBS Anderson family
6 B0034 Amp RBS
7 J23109 Amp Promoter Anderson family
8 B0032 Amp RBS Anderson family
9 R0011 Amp LacI promoter
10 B0015 Amp, Kan Double terminator
11 R0010 Amp LacI
12 E0422 Amp YFP generator
13 E0240 Amp GFP generator
14 E0040 Amp GFP
15 I13401 Amp GFP + Double terminator

Procedure:

  • Add 1 ul of DNA to 33 ul of competent cells
  • Put the tubes on ice for 10 min
  • Warm the tubes in a water bath at 37º C for 5 min.
  • Add 250 ul of LB medium
  • Incubate the cells 1 hour at 37ºC, shake at 160 rpm
