Digestion
Yuli and Hao
pSip-1::eGFP, pRab-7::eGFP, pMec-7::eGFP, unc-54 3'UTR
Cut using XbaI and SpeI. Incubated at 37 °C for 1 hour. Enzymes inactivated at 80 ° C for 20 mins.
Ligation
Hao and Yuli
pSip-1::eGFP, pRab-7::eGFP, pMec-7::eGFP, unc-54 3'UTR
Ligated using T4 ligase.
