I did another restriction digest of I0500-B0034-MalE since the restriction digest last night failed. This time I tried with EcoRI and SpeI. The restriction digest worked and Colony 10 will be sent down for sequencing tomorrow. The plates from last night did not grow either. So I retried the construction by retrying the restriction digest. I left the digest overnight and will plate tomorrow.
