Start of 6 liquid cultur from the plates of PHL1273 of yesterday :
3 at 28°C and a shaking speed of 200 rpm, 3 at 28°C and a shaking speed of 50rpm
Digestion (with Roche enzyme) :
- Curli : E+S
- Phasin, 22B (clones 1,2,3), 24M (clones 1,2,3) : X+P
- negative witness without enzyme for Curli and Phasin
The Curli and Phasin extraction were contaminated and it destroy the DNA during the digestion. We will make new extractions.
Result of the transformation :
- 8C/14K : 1 clone
- 18A/24A : 1 clone
- 4O/10L : nothing
- L3/18A (L4) :
- 12/13 :
- pha A/pha B :
