Transformation products

• XL1B: BglII-Mnt-pGEM: 14 blue colonies
• XL1B: BglII-AraC-pGEM: 13 blue colonies
• XL1B: BglII-CI-pGEM: 11 blue colonies
• BL21: RFD control: no colonies, but cannot find any protocol errors

Lovtwo transformation

• XL1B: LovTwo-pGEM: 18ul ligation
• BL21: RFP control (1:1000 dilution): 5ul

Blue-white selection to be used. Incubated @37C at 16:10

Miniprep of pBlueScript (pBS)

Gary streaked out plate containing pBS on morning of 8/17. We picked a colony, started a liquid culture in 5mL LB amp @2am on 8/18. At 6pm on 8/18, the liquid culture was harvested for miniprep to obtain stock of pBS.
